Dear Josh,
I have checked some of the single mutants myself; Kiyoshi's estimates are good. I have found, however, that starving at 0 degrees for 4 days gives very low background and therefore much prettier results. Next on the program is Gal 3 which was never done on a strictly quantitative basis which is of special importance here of course since it is a leaky mutant. Epimerase was not done on this mutant for the reason of some technical complications which I am sure that I can bypass. In about a fortnight I shall probably be able to send you data (quantitative) on Gal 3. We have pleasant surprises on our studies on Yeast 4-epimerase; there is a bond group of probably DPNH (but perhaps not the usual DPNH).
Let me hear from you!
Best regards to both of you
Sincerely
Kalckar
